淡紫灰链霉菌gCLA4对猕猴桃溃疡病预防效果及抑菌活性成分分离鉴定

中文摘要:为明确分离自黄瓜叶片的内生淡紫灰链霉菌gCLA4菌株(gCLA4)对猕猴桃溃疡病预防效果及其抗菌活性物质,通过室内离体枝条接种及田间喷淋树干的方法评价其预防效果;借助“生物活性追踪”方法,采用离子交换树脂吸附、凝胶柱层析、质谱、核磁共振波谱分析手段对gCLA4的抑菌活性物质进行分离鉴定。室内离体枝条和田间树干喷施结果表明,gCLA4菌株发酵液对猕猴桃溃疡病的防效分别为83.43%和71.07%,明显优于市场对照生防菌剂解淀粉芽孢杆菌Ba168 WP和枯草芽孢杆菌WP。室内分析鉴定显示其主要抑菌成分为链丝菌素F。皿内抑菌结果表明,链丝菌素F与商品中生菌素原药对病菌的抑制效果差异不显著,抑菌圈直径分别为20 mm和22 mm,大于gCLA4发酵滤液的抑菌圈直径16 mm。综上可见:gCLA4对猕猴桃溃疡病防控效果显著,其活性成分链丝菌素F,具有溃疡病绿色防控生物药剂开发应用的巨大价值和潜力。

Effect of Streptomyces lavendulae gCLA4 on Control of Kiwifruit Bacterial Canker and Identification of Its Antibacterial Substances
Abstract:In order to clarify the control effect and antibacterial activity of endophytic Streptomyces lavendulae gCLA4 against kiwifruit canker disease,the preventive effect of gCLA4 was evaluated by in vitro test and field drying test.The antibacterial activity compound of gCLA4 was isolated and identified by means of ion exchange resin adsorption,gel column chromatography,mass spectrometry(MS) and nuclear magnetic resonance(NMR) spectroscopy.The results showed that the control efficiency of S.lavendulae gCLA4 fermentation broth against kiwifruit canker was 83.43%(control effect test on vitro branches) and 71.07%(control effect test in kiwi orchards),respectively,which was significantly better than that of Bacillus amyloliquefaciens Ba168 WP and Bacillus subtilis WP,and slightly lower than that of Zhongshengmycin WP.Streptomycin F was identified as the main antibacterial component of S.lavendulae gCLA4.Bacteriostatic test showed that streptomycin F and Zhongshengmycin had similar inhibitory effect on Pseudomonas syringae pv.actinidiae(Psa),and the diameters of bacteriostatic zone were 20 mm and 22 mm,respectively,which were larger than that of S.lavendulae gCLA4 fermentation filtrate 16 mm.S.lavendulae gCLA4 has a significant and stable preventive effect on kiwifruit canker,which has potential development value.Moreover,the active ingredient of streptomycin F provides a basis for mining biological agents with good control effect on kiwi fruit canker disease.
