野菊抗盐株系筛选与抗盐机理初探

中文摘要:为发掘抗盐性强的野菊(Chrysanthemum indium L.)株系并揭示野菊抗盐的部分生理机制,以150 mmol/L NaCl水培作为胁迫方式,基于表型变化结合聚类分析对207个引种野菊株系进行抗盐性筛选,从得到的高、中、低抗3类中分别选择2个株系,使用霍格兰营养液(对照)、150 mmol/L NaCl(处理)两种方式对各株系进行分组处理,72 h后进行表型观察与相关生理指标测定。结果表明,引种野菊可根据抗盐性分为高、中、低抗3类,高抗株系111个,中抗株系44个,低抗株系52个。处理组与对照组在表型上差异明显,且处理组中抗盐性越弱的株系出现的胁迫症状越严重。处理组中除高抗株系的水分利用效率,高抗与中抗株系的气孔限制值,低抗株系的胞间CO2浓度高于对照组外,其余的光合指标均低于对照组。处理组的光合色素含量均低于对照组。处理组的渗透调节相关物质含量均高于对照组。处理组中,株系A到株系F的胞间CO2浓度均为先降后升趋势,表观叶肉导度、气孔限制值为先升后降趋势,丙二醛含量为上升趋势,叶绿素 a/b的趋势不具明显规律,其余指标均为下降趋势。综上,引种207个野菊株系中有111个株系对盐有较强的抗性,抗盐性较强的野菊株系在盐胁迫下较强的渗透调节能力与水分代谢能力使其表型未发生明显变化。

Screening of Salt-resistant Strains of Chrysanthemum indicum and Preliminary Study on Salt-resistant Mechanism
Abstract:In order to explore a salt-resistant C.indicum strain and reveal some physiological mechanisms of salt-resistance of C.indicum. The salt tolerance of 207 introduced C.indicum was studied by using phenotypic variation and cluster analysis under 150 mmol/L NaCl stress,the plants were divided into different groups by hoagland nutrient solution control and 150 mmol/L NaCl stress,and the phenotypes and physiological indexes were observed 72 hours later. The results showed that the salt resistance of introduced C.indicum could be divided into 3 types,including 111 high-resistant lines,44 medium-resistant lines and 52 low-resistant lines. The phenotypic difference between treatment group and control group was significant,and the less salt-resistant the strain appeared,the more severe the stress symptom was. In the treatment group,except the water use efficiency of the high-resistant and middle-resistant lines,the stomatal limitation of the high-resistant and middle-resistant lines,and the intercellular CO2 concentration of the low-resistant lines were higher than those of the control group,all other photosynthetic indexes were lower than those of the control group. The content of photosynthetic pigment in treatment group was lower than that in control group. The content of osmolytes in treatment group was higher than that in control group. In the treatment group,from strain A to strain F,the intercellular CO2 concentration decreased first and then increased,the apparent mesophyll conductance and stomatal limit value increased first and then decreased,the Malondialdehyde content increased,the chlorophyll a/b trend was not obvious,the rest of the indicators are on a downward trend.To sum up,111 out of 207 introduced lines of C.indicum have strong salt resistance,and the strong osmotic adjustment ability and water metabolism ability of the line of C.indicum have no obvious change under salt stress.
