中文摘要:为探讨甲状腺激素(T3、T4)及其合成酶( DIO2、 DIO3)对甘加藏羊生殖活动的调控,本研究以发情周期和乏情期甘加藏羊(Ovis arise)为研究对象,应用酶联免疫吸附法、实时荧光定量PCR和蛋白免疫印迹技术检测甘加藏羊发情周期和乏情期血浆中T3和T4的动态变化、下丘脑-垂体-卵巢轴(Hypothalamic-pituitary-ovarian axis, HPOA)及甲状腺组织中 DIO2、 DIO3 mRNA和蛋白的表达。结果显示:整个发情周期和乏情期血浆中T3和T4浓度均以脉冲式和波动式交替变化,T4的浓度均显著高于T3,T3的平均浓度在发情前期最高,T4的平均浓度在发情期最高,与其他各时期比较差异均显著。藏羊发情周期和乏情期HPOA和甲状腺组织中均有 DIO2 、 DIO3 mRNA及其蛋白的表达,且差异显著;垂体、卵巢、甲状腺 DIO2 mRNA及其蛋白的相对表达量均高于 DIO3 mRNA及其蛋白的相对表达量。甘加藏羊发情周期和乏情期血浆T3、T4的浓度和 DIO2、 DIO3在HPOA及甲状腺组织中的表达呈现动态变化,表明甲状腺激素及其合成酶参与调控甘加藏羊的生殖活动。 |

Dynamic Changes of T3 and T4 in Plasma and Expression of DIO2 and DIO3 in HPOA Axis of Ganjia Tibetan Sheep during Estrus Cycle and Anestrus |

Abstract:In order to explore the regulation of TH(T3, T4) and thyroid hormone synthase( DIO2, DIO3) on the reproductive activities of Ganjia Tibetan sheep , the Ganjia Tibetan sheep during the estrus cycle and anestrus were selected as the research object, enzyme-linked immunosorbent assay, real-time fluorescent quantitative PCR and western blotting technology were used to detect the dynamic changes of T3 and T4 in the plasma of Ganjia Tibetan sheep during the estrus cycle and anestrus, and the relative expression of DIO2, DIO3 mRNA and protein in the HPOA axis and thyroid tissue. The results revealed that the plasma content of T3 and T4 in the entire estrus cycle and the anestrus changed alternately in a pulsing and fluctuating manner. T4 was significantly higher than that of T3. The average concentration of T3 was the highest in the proestrus, while T4 was the highest in the estrus, which were significant compared with other periods (P<0.05). The DIO2 and DIO3 mRNA and protein were all expressed in the HPOA and thyroid tissue of the Tibetan sheep during the estrus cycle and anestrus, and the differences were significant; the relative expression of DIO2 mRNA and its protein in pituitary,ovary and thyroid were higher than the relative expression of DIO3 mRNA and its protein.The content of T3, T4 in the plasma and the expression of DIO2, DIO3 in the HPOA and thyroid during the estrus cycle and anestrus of the Ganjia Tibetan sheep changed dynamically, which indicated that TH and thyroid hormone synthase were involved in the regulation of reproductive activities in the Ganjia Tibetan sheep. |
