小麦白粉菌产孢相关基因 BgtFTT1的序列特征和基因表达分析

中文摘要:旨在揭示小麦白粉菌 BgtFTT1基因的序列特征及其在白粉菌分生孢子形成过程中的表达规律,为解析14-3-3蛋白在小麦白粉菌无性生殖调控中的作用奠定理论基础。通过基于转录组数据的序列分析和PCR扩增技术克隆 BgtFTT1基因的cDNA序列,采用生物信息学和分子生物学方法分析 BgtFTT1的序列特征和分子结构,监测该基因在白粉菌产孢过程中的表达动态。结果表明, BgtFTT1完整的开放阅读框长度为891 bp,编码1个含有296个氨基酸的偏碱性亲水蛋白,预测的分子质量为34.18 ku,属于14-3-3蛋白家族。BgtFTT1含有5个蛋白激酶C磷酸化位点,不含信号肽和跨膜螺旋。BgtFTT1的二级结构主要由α-螺旋和无规则卷曲组成,三级结构呈“球状”。BgtFTT1和源自其他白粉菌的同源蛋白在进化上独立于源自兼营腐生真菌的14-3-3蛋白。在21个白粉菌菌株中,BgtFTT1存在2个氨基酸位点的变异。 BgtFTT1基因表达水平在小麦白粉菌足细胞和分生孢子梗形成阶段显著上调。

Sequence Characteristics and Expression Analysis of a Conidiation-related Gene, BgtFTT1, from Blumeria gramins f.sp.tritici
Abstract:The aim of this study is to reveal the sequence characteristics of BgtFTT1 in Blumeria graminis f.sp.tritici (Bgt) and their expression levels during Bgt conidiogenesis, which will lay theoretical basis for uncovering the role of 14-3-3 proteins involved in Bgt asexual reproduction regulation.We cloned the cDNA sequence of BgtFTT1 by sequence analysis and PCR amplification based on RNA-seq data.The sequence characteristics and molecular structure were analyzed using bioinformatics and molecular biology methods.Gene expression dynamics of BgtFTT1 during Bgt conidiation was also monitored.The results showed that the full length of open reading frame of BgtFTT1 is 819 bp.This gene encodes a protein containing 296 amino acid residues, which is an alkaline, hydrophilic protein with comparative molecular weight of 34.18 ku. BgtFTT1 belongs to 14-3-3 protein family.It carries 5 protein kinase C phosphorylation sites, while no signal peptide and transmembrane helices.Secondary structure of the protein mainly consists of α-helix and random coil.The tertiary structure displayed a globular protein.Divergent evolution between BgtFTT1 along with homologous proteins from other powdery mildews and 14-3-3 proteins from the hemibiotrophic fungi was found.Mutation occurs at two amino acid residues in the protein sequences from 21 Bgt isolates.Gene expression levels of BgtFTT1 were significantly up-regulated during foot cell and conidiophore formation of Bgt.
