中文摘要:以东方百合‘索邦’(Sorbonne)为材料,结合前期转录组数据,克隆组蛋白去乙酰化酶基因HDA1(Histone deacetylases),命名为LoSorHDA1。LoSorHDA1序列长1 518 bp,编码505个氨基酸,生物信息学预测含有一个高度保守的组蛋白去乙酰化结构域。系统进化分析表明,LoSorHDA1与拟南芥等物种的HDA1同源蛋白聚为一支,而与HDAC家族其他成员关系较远;LoSorHDA1编码的氨基酸序列与单子叶植物小果野焦(Musa acuminata)的相似度最高,而双子叶植物HDA1则聚为另一支。RT-PCR结果显示:LoSorHDA1在‘索邦’不同组织中普遍表达,其中在茎生根、嫩茎、花被片、花丝、柱头、花柱、子房中表达量较高,在下部叶和花药中表达稍弱。亚细胞定位表达结果表明,LoSorHDA1定位在细胞核和细胞质中。荧光定量PCR结果显示:LoSorHDA1在不同花发育阶段中的各个花部组织中均有不同程度的表达,在雌雄蕊中表达趋势相反,而在内外花被片中的表达趋势较为一致,说明LoSorHDA1可能参与东方百合花发育进程,这为后续LoSorHDA1的去乙酰化修饰在百合花发育中的功能研究奠定基础。 |

Cloning and Expression Analysis of Histone Deacetylase LoSorHDA1 in Lilium Oriental Hybrid‘Sorbonne’ |

Abstract:Using the Lilium oriental hybrid‘Sorbonne’as the material, combined with the previous transcriptome data, the histone deacetylase gene HDA1(Histone deacetylases) was cloned and named LoSorHDA1. The LoSorHDA1 sequence is 1 518 bp, encoding 505 amino acids with a highly conserved histone deacetylation domain. Phylogenetic analysis indicated that LoSorHDA1 was clustered into the same group with HDA1 homologous proteins of Arabidopsis and other species, but far from other members of the HDAC family.The amino acid sequence encoded by LoSorHDA1 has the highest similarity to the monocotyledonous plant(Musa acuminate), while HDA1 of dicotyledonous plants is clustered into another. The results of RT-PCR showed that LoSorHDA1 was expressed in different tissues of ‘Sorbonne’, while highly expressed in stem roots, tender stems, perianth segments, filaments, stigmas, styles and ovary, but lower expressed in lower leaves and anthers. Subcellular localization expression indicated that LoSorHDA1 was localized in the nucleus and cytoplasm. The results of real-time PCR showed that LoSorHDA1 was expressed to a certain degrees in different flower tissues at development stages.Its expression trend was opposite in the stamen and stamen, while consistentin the inner and outer perianth pieces, which indicated that LoSorHDA1 mightparticipate in the development process of oriental lily. These results laid the foundation for the functional study of de-acetylation of LoSorHDA1 in flower development. |
