中文摘要:小分子热激蛋白(small heat shock proteins, sHSP),作为一种分子伴侣,在植物逆境胁迫响应中发挥着重要的作用。本研究克隆了水稻中2个编码小分子热激蛋白的基因 Os16.9AOs16.9B Os16.9A Os16.9B编码的150个氨基酸,相似性高达99%。 Os16.9A Os16.9B的转录方向相反,2个基因的起始密码之间的距离为2.6 kb,它们的启动子均含有CAAT-box、TATA-box、GATA-box以及与热激等胁迫应答有关的顺式作用元件HSE和ACGT aterd1; Os16.9A的启动子还包含响应冷、干旱应答的顺式作用元件LTRE, 这些顺式作用元件呈不对称分布,推测这2个基因间的序列具有双向启动子的功能,且这2个基因可能受胁迫诱导表达。该研究将为进一步研究水稻 Os16.9AOs16.9B基因的表达调控及其功能提供参考。

Sequence Analysis and Function Identification of Two Small Heat Shock Protein Gene Promoters in Rice
Abstract:As a molecular chaperone, small heat shock proteins(sHSPs)plays an important role in plant stress response. In this study, two small molecular heat shock protein genes in rice,namedOs16.9A andOs16.9B were cloned,they both encoded 150 amino acids, with 99% similarity.The transcriptional direction ofOs16.9A andOs16.9B is opposite and the distance between the initial codon of the two genes was 2.6 kb.The results of promoter element analysis show that both the promoters of these two genes contained CAAT-box, TATA-box, GATA-box, as well as the cis-acting elements HSE and ACGT aterd1 which responded to stress such as heat shock.And ACGT aterd1 and so on. The promoter ofOs16.9A also contains the cis-acting element LTRE, which responds to cold and drought. These cis-acting elements are asymmetrically distributed in the promoters ofOs16.9A andOs16.9B. So it is reasonable to assume that the sequence between the two genes may have the function of bidirectional promoter and the two genes may express under stress conditions and in a particular period of plant growth. Cloning and sequence analysis of promoters ofOs16.9A andOs16.9B genes will provide a reference for further study on expression regulation and functional analysis ofOs16.9A andOs16.9B genes in rice.
