沙门氏菌胞外囊泡对肉仔鸡免疫保护效果的研究

中文摘要:探究鼠伤寒沙门氏菌(ST)胞外囊泡(OMVs)对肉仔鸡的免疫保护效果。350只1日龄健康爱拔益加雏鸡常规条件下饲养至7日龄,选择沙门氏菌感染为阴性、健康及体质量差异不显著的雏鸡320只随机等分为5组,每组8重复,每重复8只。试验设计为未感染的对照组(PBS组)、沙门氏杆菌感染组(ST组,攻毒试验用)和3个不同OMVs免疫剂量免疫组,分别注射5、10和20 μg/(d·只)的OMVs(OMV5、OMV10和OMV20)。从8日龄开始,每隔1周对OMVs免疫组分别腹腔注射对应剂量的OMVs,每次连续3 d,共免疫3次;PBS组和ST组分别注射等量PBS溶液。试验鸡30日龄时,对ST组和3个OMVs免疫组腹腔注射1×109 CFU的ST进行攻毒,PBS组注射等量PBS溶液,饲养观察5 d。结果表明:1)在8~28日龄的免疫期,与PBS组相比,OMV5和OMV10组的试验鸡日均采食量、平均日增量和饲料转化率均无显著差异,而OMV20组的试验鸡日均采食量和平均日增量显著降低、饲料转化率显著增加;3个OMVs免疫组的死亡率及免疫器官指数与PBS组均无显著差异;2)3个OMVs免疫组试验鸡的免疫器官指数与PBS组无显著差异;OMVs免疫可显著提高免疫期间血清中的溶菌酶活力、白蛋白和C3质量浓度及SOD活力;3)与ST组相比,OMVs免疫组在ST攻毒后显著消除ST感染引起的试验鸡体质量下降,显著提高ST感染鸡的存活率,尤以OMV10和OMV20免疫组效果最好。此外,10 μg/(d·只)的OMVs免疫可显著降低ST感染后血液中的IL-1β和TNFα的质量浓度,且有降低IL-6质量浓度的趋势。由此可知,适量剂量的OMVs免疫对肉仔鸡生产性能无显著影响,但可增强肉仔鸡机体免疫功能,且具有显著提高肉仔鸡抗ST感染的能力,本试验中ST-OMVs最佳免疫剂量为10 μg/(d·只)。

Effects of Salmonella Outer Membrane Vesicles on Immune Protection of Broilers
Abstract:The study was conducted to investigate the impacts of Salmonella typhimurium outer membrane vesicles on broilers. Three hundred and twenty 1-day-old AA broilers were randomly allocated in five treatments with eight replicates of eight broilers each. ST-OMVs levels were 0, 5 μg/(d·bird), 10 μg/(d·bird), 20 μg/(d·bird)(CK, ST, OMV5, OMV10, OMV20) , respectively. Growth performance, survival, immune indexes, mass and survival(after changed by ST) were determined. The results showed that OMV20 had effect on DFI, BWG and FCR, whereas between the other groups were not significant. The OMVs levels had no effect on survival and immune indexes. OMV immunization could significantly increase LZM, ALB, C3 and SOD activity in serum during immunization. After it was changed by ST, compared with the CK group, the ST group significantly decreased body weight and survival. In addition, immunization with 10 μg/(d·bird) OMV significantly reduced the levels of IL-1β and TNFα in blood after ST infection and decreased the IL-6 level. The results showed that the appropriate dose of OMVs immunization had no significant effect on broiler performance, but it could enhance the body immune function of broilers, and had a significant increase in ability of broilers to resist ST infection. The best immunization dose of ST-OMVs in this experiment was 10 μg/(d·bird).
