中文摘要:旨在建立羊蜱蝇的分子生物学鉴定方法,从新疆南疆库车县的绵羊体表采集样品,形态学鉴定初步确定为羊蜱蝇,随后进行羊蜱蝇12S rDNA、16S rDNA和 18S rDNA基因的扩增和测序,将所得序列进行Blast在线分析和MEGA 5.0分析。结果表明,12S rDNA序列与云南2016年提交的相似性为100%,16S rDNA序列与丹麦2007年、捷克2017年和新疆2017年提交的相似性均为99%, 18S rDNA序列与新疆2016年提交的相似性为99%。羊蜱蝇16S rDNA和 18S rDNA序列均能与GenBank数据库中已知羊蜱蝇基因序列聚类,且羊蜱蝇种内K2P遗传距离为0~2.3%,通过12S rDNA、16S rDNA和 18S rDNA基因的扩增和测序可准确鉴定羊蜱蝇。 |

Identification of Melophagus ovinus Based on 12S rDNA,16S rDNA and 18S rDNA Gene in Southern Xinjiang |

Abstract:The aim of this study is to establish the molecular biological identification method of Melophagus ovinus,samples were collected from sheep’s body surface in Kuqa County,southern Xinjiang,and identified as Melophagus ovinus by morphological identification. The 12S rDNA,16SrDNA and 18S rDNAgenes were amplified and sequenced. The results of on-line Blast analysis and MEGA5.0 analysis of the sequences showed that the similarity between 12S rDNA sequence and submitted in Yunnan in 2016 is 100%,the similarity between 16S rDNA sequence and Denmark 2007,Czech Republic 2017 and Xinjiang 2017 submitted is 99%,the similarity between the 18S rDNAsequence and submitted in Xinjiang in 2016 is 99%. The sequences of 16S rDNA and 18S rDNAof the Melophagus ovinus can be clustered with the gene sequences of the Melophagusovinus known in the GenBank database,and the genetic distance of K2P in the species is 0-2.3%. 12S rDNA,16S rDNA and 18S rDNAgenes can be amplified and sequenced for accurate identification of Melophagusovinus. |
