中文摘要:为了研究陕西省紫阳县(富硒地区)山羊各组织中Fas和 Bcl-2基因表达的情况,探讨微量元素硒对Fas和 Bcl-2基因表达的影响。将年龄相仿、体质量相近,分别来自紫阳县双安镇(高硒区,n=7)和高滩镇(低硒区,n=7)的山羊作为研究对象,运用实时荧光定量PCR(qRT-PCR)技术检测心脏、肝脏、脾脏、肺脏、肾脏、肾周脂肪、背最长肌、股二头肌和淋巴组织中Fas和 Bcl-2基因mRNA的表达量。结果表明:双安镇山羊各组织Fas基因mRNA表达量由高到低依次为肝脏>心脏>肺脏>肾脏>肾周脂肪>脾脏>股二头肌>淋巴>背最长肌;高滩镇心脏和肝脏中mRNA的表达量最高,显著高于肾周脂肪、肾脏、背最长肌、股二头肌、肺脏、淋巴、脾脏(P<0.05),肝脏、肺脏(P<0.01)和肾脏(P<0.05)中的表达量均显著低于双安镇。双安镇山羊肺脏中 Bcl-2基因mRNA表达量最高,与脾脏和肾周脂肪组织差异不显著(P>0.05),但是显著高于其他组织(P<0.05);而高滩镇山羊 Bcl-2基因mRNA表达量由高到低依次为肾周脂肪>肺脏>心脏>脾脏>肾脏>淋巴>股二头肌>肝脏>背最长肌,且心脏、脾脏、肾周脂肪(P<0.01)和肺脏(P<0.05)均高于双安镇山羊。由此可见,Fas和 Bcl-2基因mRNA在紫阳县山羊各组织中均有不同程度表达,且微量元素硒上调了山羊组织中Fas基因的表达,抑制 Bcl-2基因,但是硒对Fas和 Bcl-2基因表达的影响存在组织差异性。 |

Gene Expression of Fas and Bcl-2 in Goats from Ziyang County of Shaanxi Province |

Abstract:This study was intended to ascertain the mRNA expression levels of Fas and Bcl-2 genes in some tissues of goat from Ziyang County,where is a selenium-rich region in China. Goats with similar age and weight were treated as the research objects,and they came from Shuangan Town (High selenium,n=7) and Gaotan Town (Low selenium,n=7) of Ziyang County,respectively. Real-time quantitative PCR (qRT-PCR) was used to detect the relative mRNA expression levels of Fas and Bcl-2 in heart,liver,spleen,lung,kidney,perirenal fat,longissimus muscle,biceps femoris and lymphatic. The descending order of mRNA expression levels of Fas was liver> heart> lung> kidney> perirenal fat> spleen> biceps femoris> lymphatic> longissimus muscle in the goats from Shuangan Town. In Gaotan Town,the levels of Fas mRNA in heart and liver were higher than that in other tissues (P<0.05),and the mRNA levels in liver,lung (P<0.01) and kidney (P<0.05) were lower than that from Shuangan Town. As to Bcl-2,the mRNA level in lung was the highest,but not higher than that in lymphatic and perirenal fat. What's more,the descending order of mRNA expression levels of Bcl-2 was perirenal fat> lung> heart> spleen> kidney> lymphatic> biceps femoris> heart> liver> longissimus muscle from Gaotan Town,and the of mRNA levels of Bcl-2 in heart,spleen,perirenal fat (P<0.01) and lung (P<0.05) from Gaotan Town were higher than that from Shuangan Town. Thus Fas and Bcl- 2 genes expressed differently degree and widely in various tissues of goats from Ziyang County,and the expression was affected by trace element selenium but with organizational differences. |
