[摘要]
目的 研究五加Acanthopanax gracilistylus var.gracilistylus地上部分的化学成分。方法 采用硅胶、Sephadex LH-20凝胶、ODS等柱色谱方法及制备液相色谱技术分离纯化,运用核磁共振波谱、质谱等谱学技术结合理化性质鉴定化合物结构,采用MTT法研究化合物的细胞毒活性。结果 从五加地上部分70%乙醇提取物中分离得到1个新的降羽扇豆烷型三萜3α,4α,11α-三羟基-23-降羽扇豆-20(29)-烯-28-酸(1)和4个已知羽扇豆三萜3α,11α-二羟基羽扇豆-20(29)-烯-28-酸(2)、23-氧-3α,11α-二羟基羽扇豆-20(29)-烯-28-酸(3)、23-氧-3α-羟基羽扇豆-20(29)-烯-28-酸(4)和3α-羟基羽扇豆-20(29)-烯-23,28-二酸(5)。结论 化合物1为新化合物,命名为新印象酸。活性实验结果表明化合物1对人骨肉瘤143B细胞无显著细胞毒活性。

[Abstract]
Objective To study the chemical constituents from the aerial parts of Wujia (Acanthopanax gracilistylus var. gracilistylus). Methods Silica gel, Sephadex LH-20 gel, ODS and other column chromatography methods and preparative high-performance liquid chromatography technology were used for separation and purification. Nuclear magnetic resonance spectroscopy, mass spectrometry and other spectroscopy techniques combined with physical and chemical properties were applied to identify the structures of the compounds. The in vitro cytotoxic activities of compound 1 were studied by MTT method. Results 3α,4α,11α-Trihydroxy-23-norlup-20(29)-ene-28-oic acid (1) and four known lupine triterpenoids 3α,11α-dihydroxylup-20(29)- ene-28-oic acid (2), 23-oxo-3α,11α-dihydroxylup-20(29)-ene-28-oic acid (3), 23-oxo-3α-hydroxylup-20(29)-ene-28-oic acid (4), and 3α-hydroxylup-20(29)-ene-23,28-dioic acid (5) were isolated from the 70% ethanol extract. Conclusion Compound 1, 3α,4α,11α-trihydroxy-23-norlup-20(29)-ene-28-oic acid, with no cytotoxicity to 143B cells, is a new compound named norimpressic acid.
