Abstract: OBJECTIVE To investigate the in vitro antibacterial activity of Brazilin(BN)against 10 isolates of methicillinresistant Staphylococcus aureus(MRSA)and the combinational effects of BN with amikacin and gentamicin.METHODS BN was isolated fromCaesalpinia sappan L.by column chromatography.The micro-dilution method was performed to determine minimal inhibitory concentration(MIC)of BN against MRSA,the checkboard and time-kill curve methods were performed to evaluate the combinational effects of BN with amikacin and gentamicin.RESULTS The results showed that MIC and MBC ranges of BN used alone were 16-32 and 16-128μg·ml-1,respectively.The range of fractional inhibitory concentration indices(FICI)for BN-amikacin and BN-gentamicin combinations was 0.25-2,with most strains showing synergy or addition,and no antagonistic effects.All the two combinations showed the effects of reversing resistance on most MRSA strains,which caused most of the MICs of amikacin and gentamicin reduced to 1/8-1/4in comparison with those of the two antibiotics when used alone.Time-kill analysis demonstrated synergistic effect at 24 h in two combinations,with their log10CFU·ml-1 reduction of 2.85 and 2.62,respectively.CONCLUSION Brazilin showed dose dependent in vitro anti-MRSA synergy with amikacin and gentamicin and it could reverse the resistance of MRSA strains against the two antibiotics.
