Abstract: OBJECTIVE To study the absorption and metabolism characteristics of FM0807 in the in situ single-pass perfused rat intestinal model.METHODS A perfused rat intestinal model was used for the study.FM0807 and Cur were determined at the same time by HPLC.RESULTS FM0807,which was partly metabolized to Cur,could be easily absorbed in general intestinal segments.The Pappof FM0807in four different regions of intestine of rat reduced in the order of duodenum,jejunum,ileum and colon.The result according to the analysis of variance showed that there was no significant difference(P>0.05)in the Pappof FM0807in the duodenum,jejunum and ileum.When pH was 7.4,FM0807in the duodenum and jejunum could be easily eliminated while it was difficult in ileum and colon,especially in the colon.CONCLUSION The oral administration plan of FM0807 can be optimized such as preparation of enteric-coated preparation to improve its bioavailability on account of its good property of absorption and metabolism in intestinal canal.
