黄芪通过调节环磷腺苷通路对脑缺血/再灌注大鼠神经功能的影响

摘要: 目的研究黄芪中代表成分对脑缺血/再灌注大鼠神经功能的影响及作用机制。方法 将大鼠随机分为3组:假手术组、模型组、黄芪甲苷组,黄芪甲苷组给予黄芪甲苷灌胃,假手术组和模型组给予生理盐水灌胃。黄芪甲苷组和模型组制作脑缺血模型即大脑中动脉栓塞(MCAO)模型,制造脑缺血,20 min后停止结扎恢复血流,再灌注24 h。按照改良神经功能评分的标准及方法,于造模后1 d、7 d、14 d进行神经功能评分;脑组织切片及染色,利用分析软件计算脑梗死体积;采用酶联免疫吸附实验法测定大鼠动脉血清中各酶活性变化。结果与模型组比较,黄芪甲苷组脑缺血/再灌注后7 d、14 d神经功能评分较低(P<0.05),随着脑缺血/再灌注后时间增加,黄芪甲苷组神经功能评分呈降低趋势;脑缺血/再灌注后24 h,模型组和黄芪甲苷组的脑梗死体积分别为(37.39±3.06)%、(16.38±1.67)%,差异有统计学意义(P<0.05);与模型组比较,黄芪甲苷组大鼠血清环磷酸腺苷(cAMP)、腺苷酸环化酶(AC)和蛋白激酶(PKA)活性明显升高,差异均有统计学意义(P<0.05)。结论黄芪可以调节环磷腺苷通路中的相关蛋白来减少脑缺血/再灌注大鼠脑梗死体积,改善大鼠的神经功能。

Effects of Astragalus Membranaceus on Neurological Function of Cerebral Ischemia/Reperfusion Rats by Regulating Adenosine Cyclophosphate Pathway

Abstract ObjectiveTo study the therapeutic effect and mechanism of representative components of astragalus membranaceus on neural function in rats with cerebral ischemia/reperfusion.MethodsAll rats were randomly divided into 3 groups:sham operation group,model group,astragaloside IV group.The rats in astragaloside IV group were administered astragaloside IV by gavage,and the rats in sham operation group and model group were given saline by gavage.The cerebral artery embolization (MCAO) rat models were established.After 20 minutes of cerebral ischemia,the blood flow was recovered,then the reperfusion was performed for 24 h.The neurological functional damage scores were assessed at 1,7 and 14 days after modeling.The brain tissue sections and the staining were performed,and the cerebral infarction volume was calculated by the analysis software.The enzyme activity in arterial serum of rats were determined by enzyme-linked immunosorbent assay.Results Compared with model group,the neurological function scores in astragaloside IV group were lower after cerebral ischemia/reperfusion at 1,7 and 14 days (P<0.05).With the extension of time after cerebral ischemia/reperfusion,the neurological function score of astragaloside IV group also showed a decreasing tendency.After cerebral ischemia/reperfusion 24 h,the cerebral infarction volume of model group and astragaloside IV group were (37.39±3.06)% and (16.38±1.67)%,respectively,there was statistically significant difference(P<0.05).Compared with model group,the levels of serum cyclic adenosine monophosphate (cAMP),adenylate cyclase,and protein kinase in astragaloside IV group were higher(P<0.05).ConclusionAstragalus membranaceus can reduce the area of cerebral infarction in rats with cerebral ischemia/reperfusion,and improve neurological function by regulating adenosine cyclophosphate pathway.
