碱性植烟土壤微生物群落对深耕培肥的响应特征研究

    摘要:

    为构建山地植烟土壤良好耕层,在保山市碱性植烟土壤区域设置随机区组大田试验,以深耕(30 cm)为主处理,不同培肥模式为副处理,于耕作前和烤烟收获后测定主要土壤理化性质,采用Illumina MiSeq测序技术分析微生物多样性、种群丰度、群落结构和功能类群特征,探讨深耕培肥对碱性植烟土壤质量改善效果。多样性分析结果显示,深耕配施生物有机肥和化肥处理的微生物种群的Chao指数和Shannon指数差异性不显著,但促进了微生物尤其是真菌群落组成结构差异性增强。微生物群落组成分析结果表明,与耕作前相比,化肥处理的Proteobacteria(变形菌门)、Acidobacteria(酸杆菌门)、Mortierellomycota(被孢霉门)和Basidiomycota(担子菌门)的丰度显著增加,Actinobacteria(放线菌门)和Ascomycota(子囊菌门)种群丰度显著降低;增施生物有机肥处理的Proteobacteria和Mortierellomycota的丰度显著增加,Chloroflexi、Acidobacteria和Basidiomycota的丰度显著下降;与化肥处理相比,增施生物有机肥处理的Actinobacteria、Proteobacteri和Ascomycot种群丰度显著增加,Chloroflexi、Acidobacteria和Basidiomycot丰度显著降低。BugBase表型预测和FUNGuild功能预测结果表明,深耕配施生物有机肥和化肥处理的致病性细菌和真菌丰度显著下降,显著提高土壤微生物功能类群丰度。相关性结果表明,土壤容重和有机质是影响土壤微生物群落结构的主要理化因子,深耕配施生物有机肥和化肥通过改善土壤理化性质而调节土壤细菌和真菌群落结构组成,提升有益微生物种群丰度、降低病原菌丰度。

    Abstract:

    In order to build a good arable layer of mountainous tobacco plating soil,a field experiments were conducted in the alkaline tobacco-growing area of Baoshan to study the response characteristics of soil microbial community to deep tillage and incorporated bio-organic fertilizer.Randomized field trials were conducted with deep tillage (30 cm) as the main treatment and different doses of bio-organic fertilizer as the secondary treatment.Soil physical and chemical properties,microbial abundance and diversity,community structure characteristics and functional groups of each treatment were tested and analyzed using general test methods and Illumina MiSeq sequencing techniques,respectively.Finally,the improvement effects of deep tillage and incorporated bio-organic fertilizer on alkaline tobacco-planting soil quality were discussed.The results of diversity analysis showed that there was no significant difference in Chao index and Shannon index of microbial community under the treatment of deep tillage and incorporated bio-organic fertilizer,but the difference of microbial community,especially fungal community,was enhanced.Compared with those of before cultivation,the abundance of Proteobacteria,Acidobacteria,Mortierellomycota and Basidiomycota increased significantly,while the abundance of Actinobacteria and Ascomycota were decreased significantly under chemical fertilizer treatment.In addition,the abundance of Proteobacteria and Mortierellomycota increased significantly,while the abundance of Chloroflexi,Acidobacteria and Basidiomycota decreased significantly under bio-organic fertilizer treatment.Compared with chemical fertilizer treatment,the abundance of Actinobacteria,Proteobacteri and Ascomycot populations increased significantly,while the abundance of Chloroflexi,Acidobacteria and Basidiomycot decreased significantly under bio-organic fertilizer treatment.The results of BugBase phenotypic prediction and FUNGuild functional prediction showed that the abundance of pathogenic bacteria and fungi decreased significantly,and the abundance of soil microbial functional groups increased significantly.Correlation analysis showed that soil bulk density and organic matter were the main physical and chemical factors affecting soil microbial community structure.Deep tillage and incorporated bio-organic fertilizer could regulate the composition of soil bacterial and fungal community structure by improving soil physical and chemical properties,increase the abundance of beneficial microbial population,and reduce the abundance of pathogenic bacteria.
